Abstract

การศึกษานี้มีวัตถุประสงค์เพื่อศึกษาตำแหน่งในการแสดงออก และการกระจายตัวของโปรตีน Fas และ FasL ภายในปีกมดลูกและท่อนำไข่ของกระบือปลักไทย โดยศึกษาและเปรียบเทียบความแตกต่างระหว่างระยะฟอลลิคูลาร์และระยะลูเตียลช่วงกลางของวงรอบการเป็นสัด เก็บตัวอย่างระบบสืบพันธุ์เพศเมียกระบือปลักไทยจากโรงฆ่าสัตว์ท้องถิ่น นำมาแบ่งแยกตามระยะของวงรอบการเป็นสัดโดยจำแนกจากลักษณะทางมหกายวิภาคของรังไข่ จากนั้นนำอวัยวะสืบพันธุ์เพศเมีย มาตัดแบ่งออกเป็นส่วนต่างๆ ประกอบด้วย ปีกมดลูก UTJ อิสธ์มัส แอมพูลลา และอินฟันดิบูลัม แบ่งเป็นกลุ่มตัวอย่างออกเป็น 2 กลุ่มคือ ระยะฟอลลิคูลาร์และระยะลูเตียลช่วงกลาง กลุ่มละ 5 ตัว นำตัวอย่างปีกมดลูกและท่อนำไข่ส่วนต่างๆ นำไปผ่านกระบวนการย้อมด้วยสี H&E เพื่อศึกษาทางจุลกายวิภาคศาสตร์ทั่วไป จากนั้น นำตัวอย่างทั้งหมดมาศึกษาโดยวิธีอิมมูนโนฮิสโตเคมีสำหรับการตรวจหาโปรตีน Fas/FasL และเทคนิค TUNEL assay สำหรับการเสื่อมตายของเซลล์ แล้วจึงนำมาวิเคราะห์ผลภายใต้กล้องจุลทรรศน์แสงสว่าง ผลการศึกษาพบว่า การเสื่อมตายของเซลล์ปรากฏให้เห็นอย่างชัดเจนในปีกมดลูกกระบือปลักระยะลูเตียลช่วงกลางมากกว่าระยะฟอลลิคูลาร์ ในขณะที่การเสื่อมตายของเซลล์ภายในท่อนำไข่ส่วนต่างๆ เกิดขึ้นน้อยมาก การปรากฏของโปรตีน Fas ที่เกิดขึ้นภายในปีกมดลูกและท่อนำไข่ไม่มีความแตกต่างกันเมื่อเปรียบเทียบระหว่างระยะฟอลลิคูลาร์และลูเตียลช่วงกลาง ยกเว้นในท่อนำไข่ส่วนอินฟันดิบูลัมที่พบความแตกต่างกันอย่างมีนัยสำคัญทางสถิติ (P<0.05) สำหรับการปรากฏของโปรตีน FasL พบว่า การปรากฏของ FasL ภายในท่อนำไข่ส่วน UTJ และอิสธ์มัสในระยะฟอลลิคูลาร์ มีความแตกต่างอย่างมีนัยสำคัญทางสถิติ (P<0.05) เมื่อเปรียบเทียบกับระยะลูตียลช่วงกลาง การแสดงออกของ FasL ภายใน UTJ และอิสธ์มัส อาจเกี่ยวข้องกับการทำหน้าที่ของ immune privilege ผ่านทางระบบการทำงานของ Fas-FasL ซึ่งอาจเป็นกลไกที่สำคัญส่วนหนึ่งในการทำหน้าที่ของกระบวนการกักเก็บเซลล์อสุจิภายในท่อนำไข่ของกระบือปลัก

Other Abstract (Other language abstract of ETD)

The aim of this study was to investigate the localization and distribution of Fas and FasL proteins in the uterine horns and oviducts of Thai swamp buffalo comparing between the follicular and luteal phases of estrous cycle. Female reproductive tracts of Thai swamp buffaloes were collected from local slaughterhouse and stages of estrous cycle were classified by macro-anatomical observations of both ovaries into two groups, i.e., follicular (n=5) and luteal (n=5) groups. All reproductive tracts were separated into uterine horn, UTJ, isthmus, ampulla and infundibulum, stained with H&E for histological study. Tissue samples were then examined by immunohistochemical technique for the localization of Fas/FasL proteins and TUNEL assay for apoptosis. A light microscope was used for evaluating the general histological examination, Fas/FasL immunohistochemistry and TUNEL assay. The results showed that apoptosis in uterine horn in follicular phase appeared more ubiquitous than in mid-luteal phase whereas it was rarely found in all segments of swamp buffalo oviduct at both phases. No significantly differences in the expression of Fas protein in uterine horns and oviducts except in the infundibulum (P<0.05). For the FasL protein expression, only UTJ and isthmus during follicular phase was higher than in mid-luteal phase (P<0.05). It is possible that the different FasL expression in the oviduct, especially in the UTJ and isthmus, between estrous phases might involve in the immune privilege via Fas-FasL mechanism and might be the important part to preserve spermatozoa in the sperm reservoir functions in Thai swamp buffalo.
