Abstract

วัตถุประสงค์ เพื่อศึกษาผลการยับยั้งและทำลายเชื้อแบคทีเรียพอร์ไฟโรโมแนส จิงจิวาลิสและพรีโวเทลลา อินเตอร์มีเดีย ของน้ำยาบ้วนปากผสมสารสกัดโพลีแซคคาไรด์จากเปลือกทุเรียนความเข้มข้นร้อยละ 2 โดยน้ำหนัก ในห้องปฏิบัติการ วัสดุอุปกรณ์และวิธีการศึกษา ทดสอบการยับยั้งเชื้อพอร์ไฟโรโมแนส จิงจิวาลิส สายพันธุ์ ATCC 33277และพรีโวเทลลา อินเตอร์มีเดีย สายพันธุ์ ATCC 25611 ด้วยวิธีดิส ดิฟฟิวชั่น และวิเคราะห์การทำลายเชื้อแบบไทม์คิลล์ โดยเก็บผลที่ 30 วินาที 1 และ 3 นาที ทำการตรวจนับจำนวนแบคทีเรียที่รอดชีวิต เปรียบเทียบกับน้ำยาบ้วนปากคลอร์เฮกซิดีนความเข้มข้นร้อยละ 0.1และน้ำเกลือที่ความเข้มข้นร้อยละ0.9 ผลการศึกษา เมื่อนำผลที่ได้มาวิเคราะห์ทางสถิติพบว่าน้ำยาบ้วนปากผสมสารสกัดจากเปลือกทุเรียนมีประสิทธิภาพต้านเชื้อพอร์ไฟโรโมแนส จิงจิวาลิส และพรีโวเทลลา อินเตอร์มีเดีย ได้อย่างมีนัยสำคัญทางสถิติ(p<0.001)เมื่อเปรียบเทียบกับน้ำเกลือ โดยทำให้เกิดบริเวณยับยั้งเชื้อกว้าง 11.0 และ15.5 มิลลิเมตร ตามลำดับ และเมื่อน้ำยาบ้วนปากสัมผัสกับเชื้อแบคทีเรียเป็นเวลา 30 วินาที 1 และ 3 นาที สามารถฆ่าเชื้อพอร์ไฟโรโมแนส จิงจิวาลิส และ พรีโวเทลลา อินเตอร์มีเดียได้ร้อยละ100 เมื่อเปรียบเทียบกับน้ำยาบ้วนปากคลอร์เฮก-ซิดีน พบว่าน้ำยาบ้วนปากผสมสารสกัดจากเปลือกทุเรียนทำให้เกิดบริเวณยับยั้งเชื้อน้อยกว่าคลอร์เฮกซิดีนอย่างมีนัยสำคัญ(p<0.001) แต่อย่างไรก็ตามเมื่อเปรียบเทียบความสามารถในการฆ่าเชื้อพบว่าไม่แตกต่างจากคลอร์เฮกซิดีน บทสรุป น้ำยาบ้วนปากผสมสารสกัดโพลีแซคคาไรด์จากเปลือกทุเรียนความเข้มข้นร้อยละ 2 โดยน้ำหนัก สามารถต้านเชื้อพอร์ไฟโรโมแนส จิงจิวาลิสและพรีโวเทลลา อินเตอร์มีเดีย ได้ โดยสามารถทำลายเชื้อทั้งสองชนิดได้ร้อยละ100 ภายในระยะเวลาตั้งแต่ 30 วินาที

Objective To investigate the antimicrobial effect against Porphyromonas gingivalis and Prevotella intermedia of 2% w/w concentration mouthrinse containing the extracted polysaccharide from durian fruit hulls in vitro. Materials and methods This study investigated the antimicrobial effect against Porphyromonas gingivalis ATCC 33277and Prevotella intermedia ATCC 25611 by Disc diffusion technique and Time kill analysis, recorded at 30 seconds,1 and 3 minutes by counting the colony forming unit which compare with 0.1% chlorhexidine mouthrinse and 0.9% normal saline solution. Results This study found that mouthrinse containing the extracted polysaccharide from durian fruit hulls had statistically significant antimicrobial effect against Porphyromonas gingivalis and Prevotella intermedia (p<0.001) when compare with normal saline solution. The inhibition zones indicating the two bacteria were inhibited from the mouthrinse were 11.0 and 15.5 millimeters, respectively. In time kill analysis, Porphyromonas gingivalis and Prevotella intermedia were killed at 100 % when the bacteria contacted with mouthrinse for 30 seconds, 1 and 3 minutes. The inhibition zones of mouthrinse containing the extracted polysaccharide from durian fruit hulls were less than those of chlorhexidine mouthrinse significantly (p<0.001). However, the bactericidal effect of mouthrinse containing the extracted polysaccharide from durian fruit hull was not different when compare with chlorhexidine mouthrinse. Conclusion: 2% w/w concentration mouth-rinse containing the extracted polysaccharide from durian fruit hull has an antimicrobial effect against Porphyromonas gingivalis and Prevotella intermedia 100 % at 30 seconds.
